Anther Culture

Crossbreeding is often accompanied by a long segregation process, numerous types of recombination, longer breeding time, and greater workload, and can result in the loss of recessive genes, so it is difficult to select for excellent overall traits. As an important means of haploid breeding, anther culture technology can be used to obtain haploid plants through isolated culture and then double them, which can obtain pure and stable lines in a shorter period of time and enable the expression of some recessive gene-controlled traits, i.e., shorten the breeding cycle and at the same time enrich the genetic base of the selected good lines, which is an important way of germplasm innovation and new variety selection and breeding. And, anther culture has the advantages of accelerating the purity of hybrid progeny, shortening the breeding cycle, and improving selection efficiency.

In anther culture, the genotype and growing environment of the test material, the dedifferentiation medium and additional components, culture conditions, anther development period, pretreatment, inoculation density, and doubling of haploid plants are all important factors affecting the culture effect. Lifeasible has a strict control program for each of these factors.

  • Medium types: MS, NTH, N6, B5, NH, C, R, CP, and other solid media.
  • Growth regulators: NAA, IAA, 2,4-D, KT, ZT, 6-BA.
  • Other components: activated carbon, AgNO3, vitamin C, coconut milk, nucleotides, and hydrolyzed casein.
  • Ploidy identification: morphological observation method, chromosome counting method, flow cytometry assay, stomatal guard cell chloroplast counting method.
