The protein hydrolysis process refers to the cutting of proteins into shorter fragments, i.e., peptides. Plant protein hydrolysis is a crucial step before peptide identification and analysis by mass spectrometry and is the basis for plant protein identification and characterization, biomarker discovery, etc. to proceed. Although intact proteins can be studied by mass spectrometry, protein identification is easier using smaller peptides and the coverage of identification can be improved. Therefore, the most common plant proteomics approaches tend to utilize site-specific enzymatic cleavage sites to generate smaller peptide fragments, and smaller mass peptides are more easily separated and characterized by HPLC and coupled mass spectrometry.

Lifeasible offers two methods of plant protein hydrolysis, in-gel hydrolysis (protein hydrolysis in a polyacrylamide gel matrix) and in-solution hydrolysis (protein precipitation in chloroform/methanol followed by re-dissolution in urea). Proteins separated by one- or two-dimensional gel electrophoresis can be hydrolyzed directly in-gel. In-gel hydrolysis, which is effective and reproducible, is more laborious and time-consuming. Compared to in-gel hydrolysis, the in-solution hydrolysis method is less laborious and requires less time, but in-solution hydrolysis of proteins, with low protein re-solubility, may lead to some sample loss.
