Chromatin is divided into euchromatin and heterochromatin, whereas euchromatin is in a loosely open state, allowing trans-acting factors to bind cis-regulatory elements, which regulate gene expression. Open chromatin is transposase-sensitive, and transposases can insert randomly into unprotected DNA sequences (similar to DNase), but transposases do not cleave DNA like restriction endonucleases. Assay for transposase-accessible chromatin with high throughput sequencing (ATAC-seq) utilizes a modified Tn5 transposase to cleave open DNA regions while ligating sequencing junctions. It requires a low starting volume of samples, a small amount of sequencing data, and high reproducibility, which can avoid the disadvantages of DNase-seq and MNase-seq, such as high starting volume, complicated operation, and poor reproducibility. Tn5 transposase binds easily to open chromatin.
