Microbial Diversity Sequencing Analysis

Lifeasible provides microbial diversity sequencing services for the analysis of DNA in samples provided by customers for sequencing of PCR products of specific lengths. Amplicon sequencing of bacterial (16S rDNA), fungal (18S/ITS), and functional genes can simultaneously detect dominant species, rare species, and some unknown species, and precisely analyze the composition and abundance of microorganisms at the species level, making it an important tool for current analysis of microbial diversity and differences in community composition.

  • Bacterial microbial diversity

16S rDNA sequencing. 16S rDNA is the DNA sequence encoding the small subunit rRNA of the prokaryotic ribosome. It has 10 conserved regions and 9 highly variable regions. The conserved regions reflect the affinities among biological species, and the variable regions reflect the differences among species. Sequencing of a highly variable region of 16S rDNA is used to study the diversity of community structure in microbial samples.

  • Fungal microbial diversity

18S rDNA sequencing. 18S rDNA is a DNA sequence encoding the small subunit rRNA of the eukaryotic ribosome. 18S rDNA also contains variable and conserved regions. Sequencing of a highly variable region of 18S rDNA is used to study the structural diversity of eukaryotic microbial communities in samples.

ITS1 sequencing. The ITS is divided into two regions, ITS1 and ITS2. The ITS region evolves at a rate as much as 10 times higher than that of 18S rDNA. Sequencing of ITS1 and ITS2 is used to study the structural diversity of eukaryotic microbial communities in the samples.

  • Functional gene diversity

Functional microorganisms are a group of microorganisms that have received widespread attention in nature due to their functional importance, such as nitrifying bacteria, denitrifying bacteria, ammonia-oxidizing bacteria, sulfate-reducing bacteria, and nitrogen-fixing bacteria. Functional microorganisms may be very different taxonomically but have similar genes that enable them to perform the same function, so the genes that enable these functional bacteria to perform this specific function are called functional genes, such as AOA, AOB, nifH, etc.

Functional gene sequencing, as a special class of microbial amplification and sequencing project, Lifeasible designs primers based on the gene sequences of microorganisms with specific functions in complex samples, uses the primers for PCR amplification, builds libraries, and then performs high-throughput sequencing. Functional gene sequencing can effectively study functional microbial species information in specific environments, including functional differences, taxonomy, and abundance of functional microorganisms, and combined with environmental factors at each sampling site, the relationship between environmental factors and functional bacterial community structure can be analyzed simultaneously, thus providing important guidance for microbial composition studies in complex environments.
