- Identification of pathogenic bacteria by multi-gene sequence analysis
Accurate identification of coffee pathogenic strains can be used for the screening of coffee varieties resistant to wilt. It can effectively achieve coffee wilt disease control through the selection of coffee varieties with resistance to wilt. We collected coffee wilt disease specimens and isolated several coffee pathogenic strains by the tissue isolation method. We inoculated the identified strains into isolated coffee leaves for strain pathogenicity determination. We also use partial sequences of ITS, GAPDH, CHS-1, and ACT genes to perform single gene clustering analysis for each strain, respectively. By constructing phylogenetic trees, we can effectively analyze the differences in coffee pathogenic bacteria populations.
- Breeding of coffee wilt (CWD) resistant varieties
The production and breeding of CWD resistant coffee seedlings can largely suppress the effect of disease-causing organism accumulation on the yield and quality of coffee, which is important for coffee wilt prevention and control research and treatment. We used molecular markers to identify coffee wilt resistance genes, and by identifying coffee wilt resistance genes, it is important to select disease resistant varieties.
