Immunoprecipitation of Methylated DNA

DNA methylation is an essential component of epigenetics, which plays a vital role in maintaining normal cell function, genetic imprinting, cell development, and other processes. It is one of the new research hot spots. With the increasing research on methylation, various methylation detection methods have emerged. The methylated DNA immunoprecipitation (MeDIP) method enriches genomic unmethylated or methylated fragments by immunoprecipitation with proteins containing methyl-binding structural domains or with 5mC-specific antibodies. After finally obtaining purified methylated DNA fragments, Lifeasible can be combined with existing microarray technology to detect gene methylation or with fluorescent quantitative PCR technology to quantify DNA methylation.

MeDIP coupled with real-time fluorescent quantitative PCR (MeDIP-qPCR)

qPCR is a method of adding fluorescent reagents to the PCR reaction system, detecting the PCR process in real-time using the accumulation of fluorescent signals, and finally quantitatively analyzing the unknown templates using a standard curve. qPCR can accurately quantify the concentration of the starting template in a sensitive, rapid, specific, and highly reproducible manner. Combining MeDIP and qPCR, we first enrich methylated DNA fragments with MeDIP and then detect them by qPCR. After data analysis and processing, we can obtain precise values of methylation levels of specific sites in the detected biological samples. Our method has the advantages of no need for labeled probes, high specificity, high sensitivity, high accuracy, and good reproducibility.
