Plant lncRNA-mRNA Interactions Analysis
Long noncoding RNAs (lncRNAs) refer to transcripts that are more than 200 nucleotides (nt) in length and do not contain an obvious coding sequence (CDS). The discovery of long noncoding RNAs (IncRNAs) has filled a significant gap in our understanding of posttranscriptional gene regulation in various biological processes. lncRNAs can modulate gene expression on various levels, through which biological pathways are finely tuned in plants to respond to stress and adapt to adverse conditions.
Lifeasible provides analysis services for plant lncRNA-mRNA interactions to help our customers worldwide in plant science research. Our platform is equipped with cutting-edge facilities and professional experts to support research. Here, we provide various services according to customers’ demands.
Integrative Analysis of lncRNA and mRNA Expression
- LncRNAs can cis-regulate mRNAs, and it is believed that the function of lncRNA is related to its neighboring protein-coding genes. Research on lncRNAs and co-expressed mRNAs could shed new light on the regulatory mechanisms of biological processes in plants.
- We provide conjoint analysis of lncRNA and mRNA expression, including genome-wide identification of mRNA and lncRNA, identification of differentially expressed genes and lncRNAs, identification of differentially expressed lncRNA-mRNA interaction pairs, construction of lncRNA-mRNA pathway network, verification by real-time quantification PCR (RT-qPCR), and others.
- We perform high-throughput RNA-seq on plant material provided by our customers, followed by library construction and sequencing. Reads obtained from the sequencing machines include raw reads containing adapters or low-quality bases. After the selection, the remaining clean reads were further used for assembly and gene abundance calculations. For each transcription region, an FPKM (fragment per kilobase of transcript per million mapped reads) value was calculated to quantify its expression abundance and variations.

Interacting with mRNAs

- lncRNA can be used as a molecular decoy. It indirectly regulates the expression of target genes by blocking the action of miRNAs on the target. Then, lncRNA can compete with miRNA to bind mRNA and directly regulate target genes.
- We help our customers analyze the interaction with plant lncRNA and mRNA. lncRNAs act as miRNA-competitive endogenous molecules, directly bind to mRNA, reduce miRNA binding sites, and achieve regulation of target genes.
- We provide a series of experimental methods for this analysis service, such as PAR-CLIP, HITS-CLIP, CLASH, and LIGR-seq, which are developed to define lncRNAs function and how they interact with other RNAs.
